  • Major surgery within 4 weeks of the first dose of study treatment
  • Live vaccine within 4 weeks of study treatment
  • Active or unresolved adverse events
  • Active malignancies (progressing or requiring change in treatment) in the last 24 months other than multiple myeloma

The treatment

How it is intended to work

This is how the treatment is meant to work. Whether it helps, and how safe it is, is what this trial is testing.

p300/CBP bromodomain inhibitor CCS1477. An orally bioavailable, small molecule inhibitor of the highly conserved bromodomains of the histone acetyltransferase (HAT) paralogs, p300 (E1A-associated protein p300; p300 HAT) and CREB binding protein (CBP), with potential antineoplastic activity. Upon oral administration, p300/CBP bromodomain inhibitor CCS1477 selectively and reversibly binds to the bromodomains of p300 and CBP. This disrupts the acetylation of histones and other proteins and prevents the co-activation of key transcription factors that contribute to tumor progression including the androgen receptor (AR), androgen receptor splice variants (AR-SV), hypoxia-inducible factor 1-alpha (HIF-1-alpha) and Myc proto-oncogene protein (c-Myc). The HAT paralogs p300 and CBP are key transcriptional co-activators that are essential for a multitude of cellular processes and are implicated in the progression and therapeutic resistance of certain cancers. Goals

What the study measures

Main goals

  • Objective Response Rate, defined as the percentage of patients with a confirmed partial response (PR) or better, based on IMWG criteria and assessed by Independent Review Committee (IRC) (Assessed from enrollment to date of progressive disease or death from any cause, until the end of study (up to 48 months))

Other goals

  • ORR, defined as the percentage of patients with a confirmed PR or better, based on IMWG criteria assessed by Investigator
  • Duration of Response (DoR), defined as the duration of overall response by investigator and IRC
  • Time to Response (TTR), defined as time to confirmed PR or better, by investigator and IRC
  • Very Good Partial Response (VGPR) or better rate, defined as the percentage of patients with a confirmed VGPR or better, based on IMWG criteria and assessed by investigator and IRC
  • Complete Response (CR) or better rate, defined as the percentage of patients with a confirmed CR or better, based on IMWG criteria and assessed by investigator and IRC
  • Progression Free Survival (PFS), defined as the time from enrolment until the earliest date of Progressive Disease (PD), or death due to any cause, and assessed by investigator and IRC
  • Overall Survival (OS), defined as the time from enrolment to the date of death due to any cause
  • Incidence of treatment-emergent adverse events (TEAEs), vital signs and laboratory abnormalities

Planned enrollment: 100 (estimated).
