Context: Molecular Variants and Therapeutic Impact

FLT3 variants (either internal tandem duplication or tyrosine kinase domain variants) are observed in 40% to 50% of APL cases. The presence of FLT3 variants is correlated with higher white blood cell counts and the microgranular variant (M3v) subtype.[19-23] The FLT3 variant has previously been associated with an increased risk of induction death and, in some reports, an increased risk of treatment failure.[19-25] Given the extremely high cure rates for children with APL who were treated with tretinoin and arsenic trioxide, FLT3 variants are not associated with inferior outcomes.[26]
